Absidia sp.

Sample ID: 57 Possible ID: Absidia sp.   Classification: Phylum Zygomycota Class Zygomycetes Order Mucorales Family Cunninghamellaceae   Isolation and culturing method: Isolated from moss on a wood log. The sample placed on malt extract agar + chloramphenicol. Plate was left alone for a couple of weeks and then a small volume of hyphae was excised from the agar and transferred to achieve a pure culture.   Culture appearance and growth: Fungus grew slowly. After a couple of weeks of growth, culture was white, fuzzy, and cotton-like. Edge of culture was light green. Hyphae grew above agar, pointing upward. Hyphae […]

Daldinia sp.

Sample ID: 59 Possible ID: Daldinia sp.   Classification: Phylum Ascomycota Class Ascomycetes Order Xylariales Family Xylariaceae   Isolation and culturing method: Isolated from grass tissue. The grass sample was cut in half and placed flat on malt extract agar + chloramphenicol. Plate was left alone for a couple of weeks and then a small volume of hyphae was excised from the agar and transferred to achieve a pure culture.   Culture appearance and growth: Fungus grew quickly. After one week of growth, culture was white and fuzzy. After a few more weeks of growth, culture turned grey-brown with clumps […]

Pezizomycotina species

Sample 33 Possible ID: species in Pezizomycotina   Classification: Phylum Ascomycota Subphylum Pezizomycotina   Isolation and Culturing Methods: This sample was collected from a soil sample outside of the biology building on Bucknell’s campus on March 3rd, 2018. The sample of soil was directly plated onto a MEA plate and cultured for two weeks. The sample was extracted from the soil sample plate and cultured on a new plate and grew for one week. This process was repeated an additional time to ensure its isolation.     Description of macroscopic and microscopic characteristics: Hyphae regularly septate, without clamp connections, 1-2 […]

The sample was serially diluted with water and plated on malt extract agar + chloramphenicol. Plate was left alone for a week and then a small volume of hyphae was excised from the agar and transferred to achieve a pure culture.   Culture appearance and growth: Grey/brown colored fungus grew quickly, but experienced no growth or overgrowth after […]

Aspergillus flavus

Possible ID: Aspergillus flavus   Phylum: Ascomycota Class: Eurotiomycetes Order: Eurotiales Family: Trichocomaceae   Isolation and culturing methods: Air sample taken in between Rooke Chemistry and Van Lit. for seven minutes. Air sample was taken on a plate with malt extract agar plus antibiotics. Air sample was stored at room temperature for one week in order to let collected culture grown. After one week, samples were taken and the isolates were transferred from the main plate to an new one with the same agar conditions. Absidia sp.

Possible ID: Absidia sp., potentially Absidia corymbifera   Classification: Phylum: Mucuromycotina Class: Zygomycetes Order: Mucorales Family: Mucoraceae Genus: Absidia   Isolation and culturing methods: Isolated from a plate placed under an air vent in the men’s bathroom on the third floor of the Rooke Biology building for around 10 minutes on February 19th of 2018. After being stored at room temperature for a week along with a multitude of other fungi, hyphae of the sample were taken from this plate and placed onto its own malt extract agar plate. The following week, hyphae were removed from the new plate and transferred […]

Aspergillus sp.

Possible ID: Aspergillus sp.   Classification: Phylum: Ascomycota Class: Eurotiomycetes Order: Eurotiales Family: Trichocomaceae Genus: Aspergillus   Isolation and culturing methods: Isolated from soil that was collected near senior apartment #4 on February 25th of 2018. A small amount of the soil sample was serially diluted with water and placed on several malt extract agar plates. After being stored at room temperature for a week, hyphae from serial dilution C were transferred onto two new MEA plates in order to ensure that a pure culture would be obtained.   Culture appearance and growth: Fungal species was a somewhat blueish-green fungus […]
